Specificity

This clone has been shown to specifically recognise a fusion protein of PARK7/DJ1. It also recognises a FLAG-tagged PARK7/DJ1 expressed in eukaryotic cells.
A single band is seen when Western blotting in optimised conditions with this clone. However, overloading the gel or using low dilutions can cause other bands to appear.

Recommended dilutions ICC: Use at an assay dependent dilution. ICC/IF: 1/500. IHC-Fr: Use at an assay dependent dilution. IHC-FoFr: 1/1000. WB: Use at an assay dependent dilution. Detects a band of approximately 20 kDa.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

PARK7/DJ1 is a ubiquitously expressed protein involved in various cellular processes including cell proliferation, RNA-binding, and oxidative stress. The protein has been found to colocalize within a subset of pathologic tau inclusions in a diverse group of neurodegenerative disorders known as tauopathies (Rizzu et al. 2004). Defects in PARK7/DJ1 are the cause of autosomal recessive early-onset Parkinson's disease 7 (PARK7). Parkinson's disease (PD) is a complex, multifactorial disorder that typically manifests after the age of 50 years. The disease is characterized by bradykinesia, resting tremor, muscular rigidity and postural instability. The pathology involves the loss of dopaminergic neurons in the substantia nigra and the presence of Lewy bodies (intraneuronal accumulations of aggregated proteins), in surviving neurons in various areas of the brain. PARK7 is characterized by onset before 40 years and slow progression. It has also been suggested that PARK7/DJ1 is a mitogen dependent oncogene product involved in Ras related signal transduction pathways.

Immunocytochemistry/ Immunofluorescence - PARK7/DJ1 antibody [malphaDJ-1/E2.19] (ab11251)
ICC/IF image of ab11251 stained human HeLa cells. The cells were PFA fixed (10 min), permabilised in TBS-T (20 min) and incubated with the antibody (ab11251, 1:500 dilution) for 1h at room temperature. 1%BSA / 10% normal goat serum / 0.3M glycine was used to quench autofluorescence and block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue).
