Immunohistochemical analysis of sections from ID8 induced tumours in mice, showing staining of control samples (top row) and samples from mice treated with Trabectedin (bottom row). Samples were fixed in paraformaldehyde and blocked in 5% BSA for 45 minutes at room temperature. They were incubated with ab80726 (diluted 1:100 in PBS) for 16 hours at 4°C to label PD-L1. The secondary antibody used was an Alexa Fluor® 488-conjugated goat anti-rat IgG (H + L) polyclonal (ab150157) at 1/200. The first column shows DAPI staining, the second column shows PD-L1 staining, and the final column shows DAPI and PD-L1 merged.
Ab80276 staining PD-L1 in Mouse lung tissue sections by Immunohistochemistry (IHC-P). Tissue was fixed with formaldehyde and blocked with 2% milk for 1 hour at 20°C; antigen retrieval was by heat mediation in citrate buffer and permeabilized with Triton. Samples were incubated with primary antibody (1/50 in 2% milk in TBS Triton) for 16 hours at 4°C. A biotinylated Anti- rat in Rabbit was used as the secondary antibody.
Ab80276 staining PD-L1 in Mouse pancreas tissue sections by Immunohistochemistry (IHC-P). Tissue was fixed with formalin and blocked with 2% BSA for 2 hours at 25°C; antigen retrieval was by heat mediation in citrate buffer. Samples were incubated with primary antibody (1/50 in 2% BSA) for 20 hours at 4°C. A SignalStain® Boost IHC Detection Reagent-Cell Sign (HRP, rabbit) was used as the secondary antibody.
