The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use a concentration of 1.25 µg/ml. Detects a band of approximately 34, (36), 48 kDa (predicted molecular weight: 62 kDa). Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.
Use a concentration of 4 - 8 µg/ml.
FunctionTranscriptional repressor. May promote methylation of histone H3 on 'Lys-27' by the PRC2/EED-EZH2 complex.
Tissue specificityHighest levels in heart, skeletal muscle, and pancreas, lower levels in brain, placenta, lung, liver and kidney.
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human stomach tissue labelling PHF1 with ab66275 at 4-8µg/ml. Arrows indicate positively labelled surface mucous cells and epithelial cells of the fundic gland. Magnification: 400X.
Western blot - PHF1 antibody (ab66275)
Anti-PHF1 antibody (ab66275) at 1.25 µg/ml (in 5% skim milk / PBS buffer) + 293T cell lysate at 10 µg
Secondary HRP conjugated anti-Rabbit IgG at 1/50000 dilution
