Mouse monoclonal [LPL4A.1] to Plastin L

Specificity

This antibody does not react with T or I plastin. It recognizes LPL in Calcium independent fashion. We have data to indicate that this antibody may not cross react with Mouse. However, this has not been conclusively tested and expression levels may vary in certain cell lines/tissues.

Primary antibody notes

L plastin is a leukocyte specific actin bundling protein from the fimbrin family of proteins. It has two actin binding domains and two EF hand type calcium binding domains. It gets phosphorylated on the serine residue in its headpiece region in response to a variety of inflammatory stimuli. It is hypothesized that L plastin may play a role in regulating integrin mediated adhesion in leukocytes. L plastin is expressed in majority of human cancer cell lines derived from solid tumors. Its expression is positively upregulated by testosterone in androgen receptor positive prostrate and breast cancer cells through three androgen receptor binding elements located in the upstream region of the gene.

Actin-binding protein. Plays a role in the activation of T-cells in response to costimulation through TCR/CD3 and CD2 or CD28. Modulates the cell surface expression of IL2RA/CD25 and CD69.

Tissue specificity

Detected in intestinal microvilli, hair cell stereocilia, and fibroblast filopodia, in spleen and other lymph node-containing organs. Expressed in peripheral blood T lymphocytes, neutrophils, monocytes, B lymphocytes, and myeloid cells.

Cellular localization

Cytoplasm > cytoskeleton. Cell junction. Cell projection. Cell projection > ruffle membrane. Relocalizes to the immunological synapse between peripheral blood T lymphocytes and antibody-presenting cells in response to costimulation through TCR/CD3 and CD2 or CD28. Associated with the actin cytoskeleton at membrane ruffles (By similarity). Relocalizes to actin-rich cell projections upon serine phosphorylation.
