The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use a concentration of 1 - 4 µg/ml.
Recommend PFA Fixation and Triton X-100 treatment
1/200 - 1/500. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
FunctionEndoribonuclease that cleaves single-stranded RNAs at uridylates and releases products that have 2'-3'-cyclic phosphate termini.
Tissue specificityPlacental-specific, but also associated with various malignant neoplasms.
Sequence similaritiesBelongs to the ENDOU family. Contains 2 SMB (somatomedin-B) domains.
Post-translational modificationsIt has been suggested that the active SMB domain may be permitted considerable disulfide bond heterogeneity or variability, thus two alternate disulfide patterns based on 3D structures are described with 1 disulfide bond conserved in both.
Immunofluorescent staining of Human cell line U-251MG shows positivity in nucleus but not nucleoli and cytoplasm. Recommended concentration of ab121367 1-4 µg/ml. Cells treated with PFA/Triton X-100.
Immunocytochemistry/ Immunofluorescence - Anti-PP11 antibody (ab121367)Image is courtesy of an AbReview submitted by Mr Chi-Cheng Hsieh.
Immunocytochemical immunofluorescence analysis of methanol-fixed human HEK293T cells, labelling PP11 with 121367 at a dilution of 1/100 incubated for 16 hours at 4°C in 1% BSA in PBS with 0.1% Tween20 diluent. Blocking was with 1% BSA incubated for 1 hour at 25°C. Secondary was a goat anti-rabbit polyclonal Rhodamine conjugate at 1/100.
