    Immunogen was a synthetic peptide, which represented a portion of human Pumilio homolog 2 encoded within exon 4 (LocusLink ID 23369).

Target

  • FunctionSequence-specific RNA-binding protein that regulates translation and mRNA stability by binding the 3'-UTR of mRNA targets. Its interactions and tissue specificity suggest that it may be required to support proliferation and self-renewal of stem cells by regulating the translation of key transcripts.

  • Predicted band size : 127 kDa


    ab10361 detection of human and mouse Pumilio 2 by Western blot. Samples: Whole cell lysate (50 mg) from a human monocytic (KG-1; lane 1), mouse multipotential hematopoietic (EML; lane 2) or mouse pre-B (70 Z/3; lane 3) cell lines. Antibody: ab10361 used at 0.1 mg/ml. Detection: Chemiluminescence with 3 to 5 minute exposure.
  • ICC/IF image of ab10361 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody ab10361 at 5µg/ml overnight at +4°C. The secondary antibody (green) was DyLight® 488 goat anti- rabbit (ab96899) IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
