The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
1/50 - 1/200. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
1/250 - 1/500.
Use a concentration of 1 - 4 µg/ml.
Recommend PFA Fixation and Triton X-100 treatment
FunctionCatalyzes the oxidation of sulfhydryl groups in peptide and protein thiols to disulfides with the reduction of oxygen to hydrogen peroxide. May contribute to disulfide bond formation in a variety of secreted proteins. Also seems to play a role in regulating the sensitization of neuroblastoma cells for interferon-gamma-induced apoptosis.
Tissue specificityExpressed in pancreas, brain, placenta, kidney, heart and fetal tissues. Weakly expressed in lung, liver and skeletal muscles.
Sequence similaritiesBelongs to the quiescin-sulfhydryl oxidase (QSOX) family. Contains 1 ERV/ALR sulfhydryl oxidase domain. Contains 1 thioredoxin domain.
Cellular localizationMembrane. Secreted. Cell membrane. Nucleus membrane. Seems to be predominantly targeted to the nuclear and outer plasma membrane.
