The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use at 2-5 µg/mg of lysate.
Application notesIs unsuitable for WB.
FunctionE3 ubiquitin-protein ligase involved in postreplication repair of UV-damaged DNA. Postreplication repair functions in gap-filling of a daughter strand on replication of damaged DNA. Associates to the E2 ubiquitin conjugating enzyme UBE2B to form the UBE2B-RAD18 ubiquitin ligase complex involved in mono-ubiquitination of DNA-associated PCNA on 'Lys-164'. Has ssDNA binding activity.
PathwayProtein modification; protein ubiquitination.
Sequence similaritiesBelongs to the RAD18 family. Contains 1 RING-type zinc finger. Contains 1 SAP domain. Contains 1 UBZ-type zinc finger.

Immunoprecipitation - RAD18 antibody (ab99334)
ab99334 at 3µg/ml detecting RAD18 in HeLa whole cell lysate by IP.
Lane 1: ab99334 IP at 3µg/mg of lysate
Lane 2: IP with an antibody which recognizes an downstream epitope of RAD18
Lane 3: IP control IgG
In each case, 1 mg of lysate was used for IP and 20% of the IP was loaded.
Subsequent WB detection was performed using an antibody that recognises a downstream epitope of RAD18.
Detection: Chemiluminescence with an exposure time of 3 seconds.
Two bands represent, but one or both likely represent a post-translationally modified form.
