  • Application notesIHC-P: Ready to use for 30 min at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10 min followed by cooling at RT for 20 min.
    ICC/IF: Use undiluted. Please refer to ICC/IF image below for further details.
    WB: 1/10 - 1/30. Detects a band of approximately 23 kDa (predicted molecular weight: 26 kDa). Please refer to WB image below for further details.

    Anti-Ran antibody, prediluted images

    • Human breast stained with ab31120 for 30 minutes at room temperature.
    • ab31120 staining Ran in HeLa and mouse NIH-3T3 cells, by immunofluorescence.This antibody interacts specifically well with the spindle pole-associated fraction of Ran in mitosis (see image above) and may be extremely useful in studies of mitotic functions of Ran.

      Experimental conditions:

      optimal antibody dilution: use undiluted.

      optimal fixation protocol:  PFA fixation:10 min in 3,7 % PFA  at room temperature, 5 min in PBS/ 0,1%Triton X-100,  then 3 washes in PBS.

      IF staining was performed following a standard protocol: blocking: 30 min; primary antibody: 1 hr; secondary antibody : 45 min. All incubations were at 37 °C in PBS/ 0,1% Tween containing 3% BSA.

      Ab31120 staining Ran in Hela and NIH-3T3 cells, by western blotting. This antibody worked very well with both protein extracts, although some non-specific bands were occasionally seen using concentrated  antibody.

      Optimal antibody dilution: 1/30

      WB was performed following a standard protocol: blocking: 30 min; primary antibody: 1 hr; secondary antibody: 30 min; and developed with ECL Plus (Amersham).
