The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Application notesELISA:To detect rRANTES by direct ELISA (using 100µl/well antibody solution) a concentration of at least 0.5µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant rRANTES.
Western Blot:To detect rRANTES by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant rRANTES is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.
FunctionChemoattractant for blood monocytes, memory T-helper cells and eosinophils. Causes the release of histamine from basophils and activates eosinophils. Binds to CCR1, CCR3, CCR4 and CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant RANTES protein induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). The processed form RANTES(3-68) acts as a natural chemotaxis inhibitor and is a more potent inhibitor of HIV-1-infection. The second processed form RANTES(4-68) exhibits reduced chemotactic and HIV-suppressive activity compared with RANTES(1-68) and RANTES(3-68) and is generated by an unidentified enzyme associated with monocytes and neutrophils.
Tissue specificityT-cell and macrophage specific.
Sequence similaritiesBelongs to the intercrine beta (chemokine CC) family.
Post-translational modificationsN-terminal processed form RANTES(3-68) is produced by proteolytic cleavage, probably by DPP4, after secretion from peripheral blood leukocytes and cultured sarcoma cells. The identity of the O-linked saccharides at Ser-27 and Ser-28 are not reported in PubMed:1380064. They are assigned by similarity.

Indirect ELISA - Anti-RANTES antibody (ab9783)Image from Tripathy D & Grammas P,J Neuroinflammation. 2009 Mar 16;6:10. Fig 5.; doi:10.1186/1742-2094-6-10; 16 March 2009, Journal of Neuroinflammation 2009, 6:10
Indirect ELISA measuring RANTES levels using ab9783.
Supernatants collected from rat cerebral cortical cells after various treatments were coated onto 96-well flat bottom plates and incubated overnight at 4°C. The plates were blocked with 1% BSA at 37°C for 45 min before incubation with primary antibody (1/1000) in bicarbonate buffer overnight at 4°C. An HRP-conjugated secondary IgG (1/1000) was added and incubated for 45 min at 37°C, in the dark. The reaction was developed by adding o-phenylene diamine H2O2 for 20 min. Optical density was measured at 450 nm.
