Pre-activation is required for enzymatic assays. Please dilute Human MMP-9 to 100 µg/mL in TCNB buffer (50 mM Tris, 10 mM CaCl2, 150 mM NaCl, 0.05% Brij-35 (w/v), pH 7.5), and then add p-aminophenylmercuric acetate (APMA) to a final concentration of 1 mM. Please keep the enzyme with APMA for 8-24 hour at 37°C. Please note that the optimal treatment time may need to be determined empirically. *100mM APMA stock solution should be prepared in DMSO. Please avoid adding high concentration APMA solution (>20mM) directly into the reaction as it tends to precipitate. A pre-dilution step is highly recommended.

It is recommended to reconstitute the lyophilized protein in sterile deionized water to a final concentration of 100 µg/mL. Solubilize for 30 to 60 minutes at room temperature with occasional gentle mixing. Carrier protein (0.1% HSA or BSA) is strongly recommended for further dilution and long term storage.

May play an essential role in local proteolysis of the extracellular matrix and in leukocyte migration. Could play a role in bone osteoclastic resorption. Cleaves KiSS1 at a Gly- -Leu bond. Cleaves type IV and type V collagen into large C-terminal three quarter fragments and shorter N-terminal one quarter fragments. Degrades fibronectin but not laminin or Pz-peptide.

Belongs to the peptidase M10A family. Contains 3 fibronectin type-II domains. Contains 4 hemopexin repeats.
The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme.
Processing of the precursor yields different active forms of 64, 67 and 82 kDa. Sequentially processing by MMP3 yields the 82 kDa matrix metalloproteinase-9. N- and O-glycosylated.
