Biological activityPDK4 activity was measured in a serial dilution of the recombinant protein and the minimum amount of the protein required to phosphorylate and fully inhibit PDH immunocaptured from given amount of human, bovine, rat or mouse samples was determined.
% Purified via His tag. PDK4 was purified by metal chelation column chromatography (Ni-IDA) using His Binding Columns.
PDH Kinases (PDKs) deactivate PDH by reversible phosphorylation. In humans and rodents, 4 PDK's are present in an organ-dependent manner. In all cases, PDK's are physically part of the PDH complex (binding via lipoyl domain 2 of E2), and they phosphorylate the E1 subunit at 3 specific serine residues. PDH phosphatases remove the phosphorylation and restore activity of PDH.
