Functional Studies
1 active unit/well of uPA was loaded into a 96 well plate containing assay buffer (0.1 M NaH2PO4, pH 8.0) with or without 2.5 ug of human recombinant SPINK7. The samples were loaded with a chromogenic uPA substrate (from uPA activity assay, Millipore. ECM600) and incubated for 45 min at 37C.
Then the OD (405 nm) was measured.
It seems like this recombinant protein is functional as a serine protease inhibitor and works great.
