    ab81665 is a fluorogenic substrate for ubiquitin hydrolases. Ubiquitin-AMC is an exquisitely sensitive substrate for UCH-L3 (Km = 0.039 µM) and for the Isopeptidase-T (Km = 0.17 - 1.4 µM). Ub-AMC is useful for studying ubiquitin hydrolases when detection sensitivity or continuous monitoring of activity is essential.

     

    Substrate concentrations for assay range from 0.1-1 μM. Typical enzyme concentrations for UCH-L3 are 10-100 pM and for Isopeptidase-T are 10-100 nM. Release of AMC fluorescence by UCH enzymes can be monitored using Ex380 nm and Em460 nm wavelengths, respectively.

    Ubiquitin 7-amido-4-methylcoumarin (Ub-AMC) is a sensitive fluorescence-based substrate of deubiquitinating enzymes. It is useful for monitoring deubiquitinating enzymes’ activity or screening inhibitors of deubiquitinating enzymes. The concentration of Ub-AMC used in deubiquitinating reactions is usually 0.1-1 µM. The released AMC fluorescence can be monitored using a fluorometer or plate reader with excitation and emission wavelengths at 360 nm and 460 nm, respectively.
