% SDS-PAGE. Mouse sc-tPA is reacted with an excess of Mouse PAI1 stable mutant at physiological pH. The resulting 1:1 molar complex is purified from the remaining PAI1 by affinity chromatography with immobilized antibodies to Mouse tPA.
RelevancePAI1 acts as "bait" for tissue plasminogen activator (TPA), urokinase, and protein C. Its rapid interaction with TPA may function as a major control point in the regulation of fibrinolysis.
TPA: This converts the abundant, but inactive, zymogen plasminogen to plasmin by hydrolyzing a single Arg-Val bond in plasminogen. By controlling plasmin mediated proteolysis, it plays an important role in tissue remodeling and degradation, in cell migration and many other physiopathological events.
