  • Relevance
    TaqI is a restriction endonuclease that recognizes the double-stranded sequence T/CGA and cleaves after T-1, generating fragments with 5'-cohesive ends. It carries out endonucleolytic cleavage of DNA to give specific double-stranded fragments with terminal 5'-phosphates. Taq I will only partially cleave DNA isolated from E.coli strains that have the dam methylase (dam+ strains). Taq I is inefficient in digesting single stranded DNA . Overlapping dam methylation will block cleavage by Taq I.
