  • Immunogen

    E. coli expressed human RGS 3 isoform 1 (amino acids 401-519).

  • General notes

RGS 3 (Regulator of G protein Signaling 3) is widely expressed in many tissues and cells. The protein contains an RGS domain near the carboxy terminus that serves as a GTPase activating protein (GAP). RGS 3 negatively regulates the activation of Gialpha and G alpha q. A truncated RGS 3 (RGS 3T), with its N-terminal 313 residues removed, has been shown to negatively regulate acenylyl cyclase and PLC. RGS 3T may be localized in the nucleus and RGS 3 is found in cytoplasm. Translocation to the membrane is important for the RGS proteins to interact with heterotrimeric G proteins.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
IP Use at an assay dependent concentration.
WB Use a concentration of 1 µg/ml. Detects a band of approximately 70 kDa (predicted molecular weight: 132 kDa).

Target

  • Function
Down-regulates signaling from heterotrimeric G-proteins by increasing the GTPase activity of the alpha subunits, thereby driving them into their inactive GDP-bound form. Down-regulates G-protein-mediated release of inositol phosphates and activation of MAP kinases.
  • Sequence similarities
    Contains 1 C2 domain.
    Contains 1 PDZ (DHR) domain.
    Contains 1 RGS domain.
  • Post-translational
    modifications
Phosphorylated by cyclic GMP-dependent protein kinase.
    ISGylated.
  • Cellular localization
Cytoplasm. Nucleus. Cell membrane. Long isoforms are cytoplasmic and associated with the plasma membrane. Short isoforms are nuclear.
