Target

  • RelevanceThis 140 kDa protein is the second largest subunit of RNA polymerase II, the polymerase responsible for synthesizing messenger RNA in eukaryotes. This subunit, in combination with at least two other polymerase subunits, forms a structure within the polymerase that maintains contact in the active site of the enzyme between the DNA template and the newly synthesized RNA. It is suggested that Rpb2 plays a role in enhancing transcription from prostate specific antigen genes. These genes may be linked to prostate cancer progression.

    Detection of RBP2 by Western blot and immunoprecipitation using ab10338. Sample: Whole cell lysate (50mg for Western blot; 10cm plate for IP) from 293 cells.Antibodies: ab10338 used at the indicated concentration (A) 1mg/ml (B) or 10mg/10cm plate for IP. Detection: ECL with exposure times as indicated (A) or 10 seconds (B).
  • ICC/IF image of ab10338 stained DU145 cells. The cells were 4% paraformaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab10338, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
