  • Relevance
    In the yeast, S. cerevisiae, 12 different members of a novel gene family of putative ATPases have been identified and characterised. Due to their similarity to TBP1, a human immunodeficiency tat binding protein, they have been designated the YTA family (yeast tatbinding analogues). The most outstanding feature of the YTA proteins and their relatives is that they share a region of high similarity which extends to some 300 amino acids in length. The amino acid sequences preceding the region of high similarity are less well conserved but do contain similar structural domains, i.e. coil to coil propensities, leucine zipper like structures. However, despite the highly conserved structural features among the YTA family, diverse functions have been assigned to the various family members. The putative gene products of YTA1, YTA2, YTA3 and YTA5 are all closely related to proteins known to be constituents of the human 26S proteasome. YTA2 or Rpt3 exhibits 81% overall similarity and 62% identity at the amino acid level with human TBP7 and is regarded as the yeast homologue of human proteasomal subunit TBP7. Rpt3 has been shown to be an essential gene in yeast.
