Properties

Applications

Our Abpromise guarantee covers the use of ab66041 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
IHC-FoFr Use at an assay dependent concentration. PubMed: 21040789
WB Use a concentration of 0.1 - 1 µg/ml. Predicted molecular weight: 11 kDa.

This antibody recognizes ~22 kDa (non-reduced) or 11 kDa (reduced) of S100.

IP Use a concentration of 2 - 5 µg/ml.
ELISA Use a concentration of 0.01 - 0.1 µg/ml.
IHC-P 1/300. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

Target

  • FunctionWeakly binds calcium but binds zinc very tightly-distinct binding sites with different affinities exist for both ions on each monomer. Physiological concentrations of potassium ion antagonize the binding of both divalent cations, especially affecting high-affinity calcium-binding sites.
  • Tissue specificityHighly prevalent in heart. Also found in lesser quantities in skeletal muscle and brain.
  • Sequence similaritiesBelongs to the S-100 family.
    Contains 2 EF-hand domains.
  • Cellular localizationCytoplasm.

  • Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human melanoma tissue labelling S100 with ab66041 at a dilution of 1/300 for 10 minutes at room temperature. Antigen retrieval was performed by boiling tissue sections in 10mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes.
