Description

Rabbit polyclonal to S100 - Astrocyte Marker

Specificity

This antibody recognizes all the S-100 isoforms, proteins of 21-24kDa identified as the A and B subunits of S100 protein. It stains schwannomas, ependymomas, astrogliomas, and almost all benign and malignant melanomas and their metastases. S100 protein is also expressed in the antigen presenting cells such as the Langerhans cells in skin and interdigitating reticulum cells in the paracortex of lymph nodes.

Function

Weakly binds calcium but binds zinc very tightly-distinct binding sites with different affinities exist for both ions on each monomer. Physiological concentrations of potassium ion antagonize the binding of both divalent cations, especially affecting high-affinity calcium-binding sites.

Highly prevalent in heart. Also found in lesser quantities in skeletal muscle and brain.

ab868 staining mouse brain tissue sections by IHC-P.  Sections were formaldehyde fixed and permeabilized in 0.1% Triton-X100 in blocking solution prior to blocking in 100% BioGenex Power Block Universal Blocking Reagent for 10 minutes at 22°C.  The primary antibody was diluted 1/1000 in 1% BSA in PBS pH 7.4 and incubated with the section for 1 hour at 22°C.  A HRP polymer conjugated anti-rabbit antibody was used as the secondary.   A tyramide Alexa Fluor® 594 conjugate was added and the signal was visualized using a fluorescent microscope and a rhodamine filter

Cells were paraformaldehyde fixed prior to blocking in 5% serum for 30 minutes at 25°C. The primary antibody was diluted 1/100 and incubated with the sample for 16 hours at 4°C. Alexa fluor® 488 goat polyclonal to rabbit Ig, diluted 1/400, was used as the secondary antibody.

Tissue was fixed with paraformaldehyde. Samples were incubated with primary antibody (1/500) for 12 hours at 4°C. An undiluted HRP-conjugated polyclonal was used as the secondary antibody.
