Taksony S. Senftenberg S. Niloese It shows only minor reactivity with a few members of 140 tested non-Salmonella bacteria. No reactivity with other Salmonella serogroups from group A, B, C1, C2, D, K and Y was observed (40 tested).

Applications

Application Abreviews Notes
Agglutination Use at an assay dependent concentration.

Prepare bacteria suspension in a physiological salt solution and place 10 µl on glass slide. Add 10 µl antibody solution, mix well and watch clumping. Incorporate always negative and positive controls.

ELISA 1/10.

Dilute in PBS containing 4% BSA + 0.05% Tween. Prepare bacteria suspension in PBS (5x108 bact/ml). Coat ELISA plate with 100 ul/well for 1 hour at 37°C, store overnight at 4°C. Wash 5x with tapwater with 0.05% Tween (TT). Incubate with diluted antibody, 1 hour at 37°C. Wash 5x with TT. Incubate with appropriate conjugate (anti-mouse Ig enzyme labeled antibody). Add substrate solution, incubate, stop reaction and read optical density.
