Application Abreviews Notes
IP Use at an assay dependent concentration.

By immunoprecipitation this antibody specifically binds to unglycosylated, ~75 kDa recombinant rat SCNN1B protein.

IHC-P Use at an assay dependent concentration.

  • FunctionSodium permeable non-voltage-sensitive ion channel inhibited by the diuretic amiloride. Mediates the electrodiffusion of the luminal sodium (and water, which follows osmotically) through the apical membrane of epithelial cells. Controls the reabsorption of sodium in kidney, colon, lung and sweat glands. Also plays a role in taste perception.
  • Involvement in diseasePseudohypoaldosteronism 1, autosomal recessive
    Liddle syndrome
Bronchiectasis with or without elevated sweat chloride 1
  • Sequence similaritiesBelongs to the amiloride-sensitive sodium channel (TC 1.A.6) family. SCNN1B subfamily.
  • Post-translational
Phosphorylated on serine and threonine residues.
  • Cellular localizationApical cell membrane. Apical membrane of epithelial cells.

Anti-SCNN1B antibody images

  • IHC image of SCNN1B staining in Human skeletal muscle formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab2906, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
