The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
1/50 - 1/200. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
FunctionMay play a role in cell-cell recognition and in neuronal membrane signaling. Seems to be important for the achievement of the necessary balance between dendrite elongation and branching during the elaboration of a complex dendritic arbor. Involved in the development of appropriate excitatory synaptic connectivity.
Sequence similaritiesBelongs to the SEZ6 family. Contains 2 CUB domains. Contains 5 Sushi (CCP/SCR) domains.
Cellular localizationCell membrane. Localized on dendrites and in the synaptic and postsynaptic fraction.
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Sez6 antibody (ab121347)This image is courtesy of an Abreview submitted by Hyun Jin Kim
ab121347 staining Sez6 in Mouse brain tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with paraformaldehyde and blocked with 4% serum for 1 hour at 4°C; antigen retrieval used 0.1% NaBh4. Samples were incubated with primary antibody (undiluted) for 24 hours at 4°C. An undiluted Cy3®-conjugated Goat anti-rabbit IgG polyclonal was used as the secondary antibody.
