Anti-SLC40A1 antibody images

  • ICC/IF image of ab85370 staining of SLC40A1 in rat primary hippocampal cell culture. The sections were incubated in 10% serum to block non-specific protein-protein interactions and in 0.3% triton X to permeabilise the cells. The sections were then incubated with ab85370 (1:1000) for four hours at +24°C, followed by Alexa Fluor 488 conjugated secondary antibody (green staining). Nucleus was stained using Hoechst.

    Observed band size : 73 kDa (why is the actual band size different from the predicted?)
    Additional bands at : 46 kDa,58 kDa,65 kDa. We are unsure as to the identity of these extra bands.

    Exposure time : 16 minutes

    This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 2% Bovine Serum Albumin before being incubated with ab85370 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution ab133406.
