  • Function
    Core component of the SMC5-SMC6 complex, a complex involved in DNA double-strand breaks by homologous recombination. The complex may promote sister chromatid homologous recombination by recruiting the SMC1-SMC3 cohesin complex to double-strand breaks. The complex is required for telomere maintenance via recombination in ALT (alternative lengthening of telomeres) cell lines and mediates sumoylation of shelterin complex (telosome) components which is proposed to lead to shelterin complex disassembly in ALT-associated PML bodies (APBs). Required for recruitment of telomeres to PML nuclear bodies.

  • Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human prostate carcinoma tissue labelling SMC6L1 with ab18039 at 1/1000 (1µg/ml). Detection: DAB.
  • Detection of Human SMC6 by Western Blot and Immunoprecipitation. Samples:

    A) Whole cell lysate (WCL; 10, 20 and 50 µg) from lymphoblastoid cells (TK6).

    B) Whole cell lysate (500 µg) from HeLa and 293T cells.

    Antibody: Affinity purified rabbit anti-SMC6 antibody used at 0.1 µg/ml for WB and at 5 µg/0.5 mg lysate for IP.

    Detection: Chemiluminescence with exposure times of less than 2 minutes.

    Detection of Human SMC6 by Western Blot and Immunoprecipitation. Samples:

    A) Whole cell lysate (WCL; 10, 20 and 50 µg) from lymphoblastoid cells (TK6).

  • ab18039 (1/200) staining SMC6L in HeLa cells (green). Cells were fixed in methanol, permeabilised with 0.5% Triton X100/PBS and counterstained with DAPI in order to highlight the nucleus (red). Please refer to abreview for further experimental details.

  • IHC image of ab18039 staining in human normal tonsil formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with EDTA (pH9, epitope retrieval solution 2) for 20 mins. The section was then incubated with ab18039, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
