Our Abpromise guarantee covers the use of ab101227 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
ICC/IF Use at an assay dependent concentration. PubMed: 24797668
WB Use a concentration of 1 µg/ml. Detects a band of approximately 49 kDa (predicted molecular weight: 57 kDa).

  • Function
Involved in the transcriptional regulation of genes required for mesoderm differentiation. Probably plays a role in limb pattern formation.
  • Involvement in disease
Defects in TBX5 are the cause of Holt-Oram syndrome (HOS) [MIM:142900]. HOS is a developmental disorder affecting the heart and upper limbs. It is characterized by thumb anomaly and atrial septal defects.
  • Sequence similarities
    Contains 1 T-box DNA-binding domain.
  • Cellular localization
  • Information by UniProt

  • All lanes : Anti-TBX5 antibody (ab101227) at 1 µg/ml

Lane 1 : F9 (Mouse embryonic carcinoma cell line) Whole Cell Lysate
Lane 2 : NIH 3T3 (Mouse embryonic fibroblast cell line) Whole Cell Lysate
Lane 3 : E10 Mouse Embryo Brain Tissue Lysate

Lysates/proteins at 10 µg per lane.

All lanes : Goat Anti-Rabbit IgG H&L (HRP) preadsorbed (ab97080) at 1/5000 dilution

    Developed using the ECL technique.

    Performed under reducing conditions.

    Predicted band size: 57 kDa
Observed band size: 49 kDa (why is the actual band size different from the predicted?)

    Exposure time: 8 minutes

The predicted molecular weight of TBX5 is 57 kDa (SwissProt), however we expect to observe a banding pattern at 49 kDa. Abcam welcomes customer feedback and would appreciate any comments regarding this product and the data presented above.
