Our Abpromise guarantee covers the use of ab60727 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
Flow Cyt Use 1µg for 106 cells. ab170190-Mouse monoclonal IgG1, is suitable for use as an isotype control with this antibody.
WB Use at an assay dependent concentration. Detects a band of approximately 80 kDa (predicted molecular weight: 71 kDa).
ELISA Use at an assay dependent concentration.

  • FunctionTranscription factor that binds to the immunoglobulin enchancer Mu-E5/KE5-motif. Binds to the E-box present in the somatostatin receptor 2 initiator element (SSTR2-INR) to activate transcription (By similarity). Preferentially binds to either 5'-ACANNTGT-3' or 5'-CCANNTGG-3'.
  • Tissue specificityExpressed in adult heart, brain, placenta, skeletal muscle and to a lesser extent in the lung. In developing embryonic tissues, expression mostly occurs in the brain.
  • Involvement in diseaseDefects in TCF4 are a cause of Pitt-Hopkins syndrome (PTHS) [MIM:610954]. PTHS is a rare syndromic encephalopathy characterized by severe psychomotor delay, epilepsy, daily bouts of diurnal hyperventilation starting in infancy, mild postnatal growth retardation, postnatal microcephaly, and distinctive facial features. Since most hitherto reported cases have been sporadic, with males and females equally affected, PTHS is regarded as an autosomal dominant condition.
  • Sequence similaritiesContains 1 basic helix-loop-helix (bHLH) domain.
  • Domainthe 9aaTAD motif is a transactivation domain present in a large number of yeast and animal transcription factors.

  • Anti-TCF4 antibody (ab60727) + A549 cells

Predicted band size : 71 kDa
Observed band size : 80 kDa (why is the actual band size different from the predicted?)
Additional bands at : 40 kDa. We are unsure as to the identity of these extra bands.
  • Overlay histogram showing HeLa cells stained with ab60727 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab60727, 1µg/1x106 cells) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

Product Wall

I can confirm that the product ab60727 is against the TCF4 protein NP_001077431.1 (
http://www.uniprot.org/uniprot/P15884). It seems like the author of the paper have listed the wrong abID. I will be removing the publication from datasheet.
