Specificityab80754 recognises human Tumour endothelial marker 7 (TEM7), a 55.8 kDa protein belonging to the plexin family.
TEM7 is strongly expressed in endothelial cells from colorectal cancer and from primary cancers of the lung, liver, brain, breast and pancreas. High expression of TEM7 is associated with metastasis and poor survival of patients with osteogenic sarcoma.
Studies have indicated the existence of least four isoforms of TEM7 which include membrane bound, secreted and intracellular forms.
TEM7 is not detectable in endothelial cells from normal tissues.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use a concentration of 0.5 - 1 µg/ml. Detects a band of approximately 56 kDa (predicted molecular weight: 56 kDa).
Use a concentration of 2.5 µg/ml. Do not perform antigen retrieval.
FunctionPlays a critical role in endothelial cell capillary morphogenesis.
Tissue specificityDetected in endothelial cells from colorectal cancer, and in endothelial cells from primary cancers of the lung, liver, pancreas, breast and brain. Not detectable in endothelial cells from normal tissue. Expressed in fibrovascular membrane with increased expression in individuals with proliferative diabetic retinopathy.
Sequence similaritiesBelongs to the plexin family.
Cellular localizationCytoplasm; Secreted and Cell membrane. Cell junction > tight junction. Localized predominantly at the tight junctions of vascular endothelial cells and to a lesser extent at the luminal surface of vascular endothelial cells.
