Applications

Our Abpromise guarantee covers the use of ab81679 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 µg/ml. Predicted molecular weight: 134 kDa. Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.
ELISA Use at an assay dependent concentration.

ELISA titre using peptide based assay: 1:62500.

Target

  • FunctionPlays a role in tight junctions and adherens junctions.
  • Tissue specificityThis protein is found in epithelial cell junctions. Isoform A1 is abundant in the heart and brain. Detected in brain and skeletal muscle. It is present almost exclusively in normal tissues. Isoform C1 is expressed at high level in the kidney, pancreas, heart and placenta. Not detected in brain and skeletal muscle. Found in normal as well as in most neoplastic tissues.
  • Involvement in diseaseDefects in TJP2 are involved in familial hypercholanemia (FHCA) [MIM:607748]. FHCA is a disorder characterized by elevated serum bile acid concentrations, itching, and fat malabsorption.
  • Sequence similaritiesBelongs to the MAGUK family.
    Contains 1 guanylate kinase-like domain.
    Contains 3 PDZ (DHR) domains.
    Contains 1 SH3 domain.
  • Cellular localizationCell junction > adherens junction. Cell membrane. Cell junction > tight junction. Nucleus. Also nuclear under environmental stress conditions and in migratory endothelial cells and subconfluent epithelial cell cultures.

    Secondary
HRP conjugated anti-Rabbit IgG at 1/50000 dilution

Predicted band size : 134 kDa
Observed band size : 100 kDa (why is the actual band size different from the predicted?)
Additional bands at : <40 kDa. We are unsure as to the identity of these extra bands.
