Application Abreviews Notes
WB Use at an assay dependent concentration. Preliminary experiments gave an approx 85kDa band in Mouse fetal Brain lysates after 0.3µg/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 55.9kDa according to NP_775567.2. The 85kDa band was successfully blocked by incubation with the immunizing peptide. We would appreciate any feedback from people in the field.
IHC-P Use a concentration of 0.5 - 2 µg/ml.

Note=Naturally occurring variants of TPH2 with impaired enzyme activity could cause deficiency of serotonin production and result in an increased risk of developing behavioral disorders.

Anti-TPH2 antibody images

  • IHC-P image of THP2 staining on mouse sections of Substantia Nigra using ab121020 (1:800). The sections were deparaffinized and subjected to heat mediated antigen retrieval using citric acid. The sections were them blocked using 1% BSA for 10 mins at 21°C. ab121020 was diluted 1:800and incubated with the sections for 2 hours at 21°C. The secondary antibody used was rabbit polyclonal to goat IgG conjugated to HRP.

  • ab121020 staining TPH2 by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) of Mouse Brain stem shows staining of Raphe nucleii serotonergic neurons: Left = sagittal section, Right = coronal section. Antibody diluted to 0.5 μg/ml.
