    Mouse monoclonal [CH1] to Tropomyosin
  • SpecificityThis antibody is specific for human striated muscle forms of tropomyosin, including cardiac alpha tropomyosin and skeletal muscle forms of tropomyosin. This rod-shaped protein is associated with contractile tissue, where it regulates the Ca 2+ -dependent interaction between actin and myosin. This antibody reacts with human striated muscle forms of tropomyosin. It also cross-reacts with chicken cardiac muscle. This antibody does not cross-react with smooth muscle or non-muscle isoforms of tropomyosin.

Anti-Tropomyosin antibody [CH1] images

  • Anti-Tropomyosin antibody [CH1] (ab7786) at 1/200 dilution + Human skeletal muscle tissue lysate - total protein (ab29330) at 10 µg

    Goat polyclonal to Mouse IgG - H&L - Pre-Adsorbed (HRP) at 1/3000 dilution

    Observed band size : 37 kDa (why is the actual band size different from the predicted?)
    Additional bands at : 40 kDa. We are unsure as to the identity of these extra bands.
  • Overlay histogram showing HeLa cells stained with ab7786 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab7786, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells ) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in HeLa cells fixed with 4% paraformaldehyde (10 min)/permeabilized in 0.1% PBS-Tween used under the same conditions.
