  • Target

    • FunctionThought to form a receptor-activated non-selective calcium permeant cation channel. Probably is operated by a phosphatidylinositol second messenger system activated by receptor tyrosine kinases or G-protein coupled receptors. Activated by diacylglycerol (DAG) in a membrane-delimited fashion, independently of protein kinase C. Seems not to be activated by intracellular calcium store depletion.
    • Tissue specificityExpressed primarily in placenta, lung, spleen, ovary and small intestine. Expressed in podocytes and is a component of the glomerular slit diaphragm.
    • Involvement in diseaseDefects in TRPC6 are the cause of focal segmental glomerulosclerosis type 2 (FSGS2) [MIM:603965]. A renal pathology defined by the presence of segmental sclerosis in glomeruli and resulting in proteinuria, reduced glomerular filtration rate and edema. Renal insufficiency often progresses to end-stage renal disease, a highly morbid state requiring either dialysis therapy or kidney transplantation.

      Lane 1:Anti-TRPC6 antibody, (1:200).

      Lane 2: Anti-TRPC6 antibody, preincubated with the control peptide antigen. 

    • ab12249 staining TRPC6 in mouse Olfactory tissue section by Immunohistochemistry (PFA perfusion fixed frozen sections). Mice were anesthetized with 20% chloral hydrate (2 mg/g body weight), perfused transcardially with 0.9% saline followed by 4% paraformaldehyde in 0.1 M phosphate buffer (PB). The olfactory organs were dissected and postfixed in the same fixative for 15 min to overnight. Cryosections were rinsed in 0.1 M phosphate buffered saline (PBS), blocked in blocking solution containing 1% BSA, 3% normal donkey serum, and 0.3% Triton X-100 in PBS for 2 hours, and then incubated in the primary antisera overnight to 2 days. An Alexa Fluor® 568 conjugated anti rabbit IgG was used for 2 hours as secondary. ab12249 stains red in image shown.
