Function
Nonselective, voltage-independent cation channel mediating sodium and calcium ion influx in response to oxidative stress. Extracellular calcium passes through the channel and acts from the intracellular side as a positive regulator in channel activation. Activated by ADP-ribose, nicotinamide adenine dinucleotide (NAD(+)), reactive nitrogen species and arachidonic acid. Inactivated by intracellular ATP. Confers susceptibility to cell death following oxidative stress. Isoform 2 does not seem to be regulated by ADPR. Has ADP-ribose pyrophosphatase activity. -
Tissue specificity
Highly expressed in brain and peripheral blood cells, such as neutrophils. Also detected in bone marrow, spleen, heart, liver and lung. Isoform 2 is found in neutrophil granulocytes. -

Developed using the ECL technique.
Predicted band size: 171 kDa
Exposure time: 3 minutes -
IHC image of ab11168 staining in rat brain formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab11168, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times. -
Detection of human TRPM2 by western blot of immunoprecipitates. Samples: Membrane Prep (0.5 or 1.0mg per IP reaction; 20% of IP loaded) from Jurkat cells. Antibodies: Affinity purified rabbit anti-TRPM2 antibody A300-414A (lot A300-414A-3) used for IP at 6ug per reaction. TRPM2 was also immunoprecipitated by rabbit anti-TRPM2 antibody A300-413A. For blotting immunoprecipitated TRPM2, A300-414A was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.
