IHC-P Use a concentration of 0.125 µg/ml.
ICC/IF Use a concentration of 1 µg/ml.
ELISA Use a concentration of 0.5 - 2 µg/ml. This allows the detection of 0.2 - 0.4 ng/well of recombinant human TSLP, using 100µl/well antibody solution.
WB Use a concentration of 0.1 - 0.2 µg/ml. Detects a band of approximately 28 kDa (predicted molecular weight: 18 kDa). Detection limit for recombinant hTSLP is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

    The 28 kDa band observed is comparable to the molecular weight seen with other commercially available antibodies to TSLP.
  • ab47943  staining TSLP in human prostate tissue section by Immunohistochemistry (Formalin-PFA fixed paraffin-embedded sections). Tissue underwent heat mediated antigen retrieval in sodium citrate buffer (pH 6.0). The primary antibody was used at 0.125 ug/ml and incubated with sample at 4°C overnight. A HRP-labeled polymer detection system was used with a DAB chromogen.

  • ICC/IF image of ab47943 stained HepG2 cells. The cells were 4% formaldehyde (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab47943, 1µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899 Dylight 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
