    Probably involved in intracellular signal transduction by being involved in the initiation of type I IFN signaling. Phosphorylates the interferon-alpha/beta receptor alpha chain.

    Defects in TYK2 are the cause of protein-tyrosine kinase 2 deficiency (TYK2 deficiency) [MIM:611521]; also known as autosomal recessive hyper-IgE syndrome (HIES) with atypical mycobacteriosis. TYK2 deficiency consists of a primary immunodeficiency characterized by recurrent skin abscesses, pneumonia, and highly elevated serum IgE.

    Lanes 1 - 4: Merged signal (red and green). Green - ab57678 observed at 135 kDa. Red - loading control, ab181602, observed at 37 kDa.

    ab57678 was shown to specifically react with TYK2 when TYK2 knockout samples were used. Wild-type and ProteinX knockout samples were subjected to SDS-PAGE. ab57678 and ab181602 (loading control to GAPDH) were diluted 1 μg/mL and 1/10 000 respectively and incubated overnight at 4°C. Blots were developed with Goat anti-Mouse IgG H&L (IRDye® 800CW) preadsorbed (ab216772) and Goat Anti-Rabbit IgG H&L (IRDye® 680RD) preadsorbed (ab216777)

    secondary antibodies at 1/10 000 dilution for 1 h at room temperature before imaging.

  • TYK2 antibody (ab57678) used in immunohistochemistry at 3ug/ml on formalin fixed and paraffin embedded human lymphoma.
