The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Application notesIHC-P: 1/100 - 1/500.
Epitope exposure is recommended.
Epitope exposure with citrate or tris-EDTA pH9.0 buffer will enhance staining.
Likely to work with frozen sections.
Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
FunctionNegative regulator of DNA damage repair which specifically deubiquitinates monoubiquitinated FANCD2. Also involved in PCNA-mediated translesion synthesis (TLS) by deubiquitinating monoubiquitinated PCNA. Has almost no deubiquitinating activity by itself and requires the interaction with WDR48 to have a high activity.
Sequence similaritiesBelongs to the peptidase C19 family.
Developmental stageCell cycle-regulated. Highest level during S phase.
Post-translational modificationsAutocatalytic cleavage of USP1 following UV irradiation inactivates it leading to an increase in ubiquitinated PCNA, recruitment of POLH and translesion synthesis. Ubiquitinated; leading to its subsequent proteasomal degradation. Phosphorylated upon DNA damage, probably by ATM or ATR.
