Applications

Our Abpromise guarantee covers the use of ab27794 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 - 4 µg/ml. Detects a band of approximately 34 kDa (predicted molecular weight: 40 kDa).Can be blocked with Wnt2 peptide (ab54079).
ICC/IF Use a concentration of 5 µg/ml.

Target

  • FunctionLigand for members of the frizzled family of seven transmembrane receptors. Probable developmental protein. May be a signaling molecule which affects the development of discrete regions of tissues. Is likely to signal over only few cell diameters.
  • Tissue specificityExpressed in brain in the thalamus, in fetal and adult lung and in placenta.
  • Sequence similaritiesBelongs to the Wnt family.
  • Cellular localizationSecreted > extracellular space > extracellular matrix.
  • Information by UniProt
  • Database links
  • FormThe WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis.

  • Anti-Wnt2 antibody (ab27794) at 2 µg/ml + Rat kidney tissue lysate. at 30 µg

Predicted band size : 40 kDa
Observed band size : 34 kDa (why is the actual band size different from the predicted?)
  • ICC/IF image of ab27794 stained MCF7 cells. The cells were 4% PFA fixed (10mins) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab27794, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899 Dylight 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

  • ab27794 staining Wnt2 in murine coronal tissue by Immunohistochemistry (PFA perfusion fixed frozen sections).
    (i) Developing aorta.
(ii) and (iii) Medio-lateral regions of the developing mesencephalon.
Abbreviations: BC, blood cells; AA, aorta; VM, ventral midbrain.
