Target

  • FunctionLigand for members of the frizzled family of seven transmembrane receptors. Can activate or inhibit canonical Wnt signaling, depending on receptor context. In the presence of FZD4, activates beta-catenin signaling. In the presence of ROR2, inhibits the canonical Wnt pathway by promoting beta-catenin degradation through a GSK3-independent pathway which involves down-regulation of beta-catenin-induced reporter gene expression. Suppression of the canonical pathway allows chondrogenesis to occur and inhibits tumor formation. Stimulates cell migration. Decreases proliferation, migration, invasiveness and clonogenicity of carcinoma cells and may act as a tumor suppressor. Mediates motility of melanoma cells. Required during embryogenesis for extension of the primary anterior-posterior axis and for outgrowth of limbs and the genital tubercle. Inhibits type II collagen expression in chondrocytes.
  • Tissue specificityExpression is increased in differentiated thyroid carcinomas compared to normal thyroid tissue and anaplastic thyroid tumors where expression is low or undetectable. Expression is found in thyrocytes but not in stromal cells (at protein level).

  • ab72583 staining Wnt5a in human tonsil tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde; antigen retrieval was by heat mediation. Samples were incubated with primary antibody (1/100) for 32 minutes at 37°C. An undiluted biotin-conjugated goat anti-rabbit IgG polyclonal was used as the secondary antibody.

  • ab72583 staining Wnt5a in Mouse rib tissue by Immunohistochemistry (Frozen sections). The sections were PFA-fixed prior to blocking with 20% Serum for 1 hour at 19°C. The primary antibody was diluted 1/100 and incubated with the sample for 16 hours at 4°C. An Alexa Fluor® 647-conjugated anti-Rabbit polyclonal was used as the secondary antibody, diluted 1/200. DAPI was used for the blue counterstain.

  • Immunohistochemical analysis of mouse hippocampus tissue, staining Wnt5a with ab72583.

    Tissue was fixed with paraformaldehyde before being sectioned on a microtome. The sections were permeabilized in Triton X-100 in PBS for 1 hour at room temperature. Samples were incubated with primary antibody (5 µg/ml in blocking solution) at 4°C overnight. A Cy3-conjugated donkey anti-rabbit IgG (1/200) was used as the secondary antibody.
