Our Abpromise guarantee covers the use of ab12132 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
ChIP: Use at an assay dependent dilution. (PubMed: 20106920)
ICC/IF: Use a concentration of 1 - 5 µg/ml.
WB: 1/500 - 1/1000.Detects a band of approximately 63 kDa (predicted molecular weight: 49.7 kDa).(For optimal results, primary antibody incubations should be performed at 4°C.)
EMSA: Use at an assay dependent concentration. (PubMed: 22210889)
Multifunctional transcription factor that exhibits positive and negative control on a large number of cellular and viral genes by binding to sites overlapping the transcription start site. May play an important role in development and differentiation. The function of YY1 as an activator or a repressor is specified by the presence of other proteins. For example it acts as a repressor in absence of adenovirus E1A protein but as an activator in its presence.
Belongs to the YY transcription factor family.
Contains 4 C2H2-type zinc fingers.
Nucleus matrix. Associated with the nuclear matrix.

Western blot - YY1 antibody (ab12132)

Predicted band size : 49.7 kDa
Detection of YY1 by Western blot analysis. YY1 can be detected in nuclear extracts derived from Raji cells using ab12132 (lane 1) at a 1:1000 dilution. The signal corresponding to nuclear YY1 is eliminated by adding the immunizing peptide to the reaction mixture (lane 2).
Immunocytochemistry/ Immunofluorescence-YY1 antibody(ab12132)

ICC/IF image of ab12132 stained HeLa cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab12132, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
