IP Use at 10 µg/mg of lysate.

Target

  • FunctionMay play a role in the establisment and maintenance of neuronal transmission and plasticity via its ubiquitin ligase activity. E3 ubiquitin ligases accept ubiquitin from an E2 ubiquitin-conjugating enzyme in the form of a thioester and then directly transfer the ubiquitin to targeted substrates.
  • Tissue specificityHighly expressed in the brain, with higher expression during development than in adult. Expressed also in mammary glands, testis, colon and kidney.
  • PathwayProtein modification; protein ubiquitination.
  • Sequence similaritiesContains 1 RING-type zinc finger.
  • DomainThe RING-type zinc finger domain is required for E3 ligase activity.
  • Post-translational
    modifications
Phosphorylated upon DNA damage, probably by ATM or ATR.
  • Cellular localizationEndosome membrane. Lysosome membrane. Cell junction > synapse > presynaptic cell membrane. Present in presynaptic plasma membranes in neurons.

Lane 1 : HeLa cell lysate at 50 µg
Lane 2 : HeLa cell lysate at 15 µg
Lane 3 : HeLa cell lysate at 5 µg
    Lane 4 : 293T cell lysate at 50 µg

    Developed using the ECL technique

Predicted band size : 24 kDa

    Exposure time : 3 minutes
  • HeLa whole cell lysate (1 mg) was immunoprecipitated with 10 µg ab86778 (lane 1) or control IgG antibody (lane 2). 20% of the immunoprecipitate was subjected to Western blotting and detected with ab86778 at 1 µg/ml. Bands were detected by chemiluminescence with an exposure time of 30 seconds.
