      The 10kD Spin Columns (ab93349) are disposable ultrafiltration devices for the concentration or deproteinization of biological samples.


      They can be used in either a swing bucket or fixed angle rotors which accept 2.0 mL centrifuge tube at maximum speed 10000 x g (5-10 mins). The vertical membrane design and thin channel filtration chamber minimizes membrane fouling and provides high speed concentration, even laden solutions. It is specifically designed with low internal surface and membrane area in order to achieve superior recovery from dilute solution.


      Maximum initial sample volume is 500µL. The dead stop volume is approximately 20-25µL.

    • Notes

      This product is manufactured by BioVision, an Abcam company and was previously called 1997 10K Spin Column. 1997-25 is the same size as the 25 unit size of ab93349.

      If working with cell or tissue lysates, we generally recommend the TCA Sample Deproteinization Kit or using a PCA/KOH Deproteinization protocol. If it is critical to use a spin column with a cell or tissue lysate then, to avoid blocking the filter, first centrifuge the lysate samples at ~14,000 rpm for 10-20 mins to pellet debris, and then use the resulting supernatant with the spin column.

    Please kindly recommend for L-Carnitine Assay.(ab83392) 1. What is the type of 96 well plate for use? ELISA plate? 2. Need deproteinzing sample preparation kit (ab93299) to the Cell sample and Serum sample or not. 3. Let me know to the 10kD spin Column (ab93349) protocol for serum sample and cell sample 4. Regarding protocol of Carnitine Standard Curve step. For the Colorimetric Assay: After dilute add standard Carnitine + Assay buffer in well is 0+50 ul , 2ul+48 ul, 4ul+46 ul, ………., 10 ul+ 40 ul, Right ? For the Fluorometric Assay: Dilute 1 µl of the 100 mM Carnitine Standard with 999 µl ,Right? 5. How many well for Background control/plate? or one background control/one sample? 6. Regarding Calculation step. Please check and recommend. Example: C=Sa/Sv (nmol/ul) Sa= 8 nmol Sv= 50 ul If dilute deproteinized serum samples 10x C= (8/50)*10 nmol/ul C= 1.6 nmol/ul

    1. Recommended plates would be black with clear bottoms for fluorometric assay but for colorimetric assay clear 96 well plates. 2. Yes, the samples have to be deproteinized for use with L-Carnitine Assay Kit. 3. The sample must be loaded on the column, spun at 10,000g for 5 minutes and then the elute can be used for the assay. 4. Yes, the calculations are correct for both standards. 5. Perform background control if high levels of acyl -CoA’s or free Coenzyme A are suspected to be in your samples. Choline in samples will give a positive signal but is present at ~10% of the Carnitine concentration. 5. Yes, the calculation is correct.
