FunctionAdapter protein implicated in the regulation of a large spectrum of both general and specialized signaling pathways. Binds to a large number of partners, usually by recognition of a phosphoserine or phosphothreonine motif. Binding generally results in the modulation of the activity of the binding partner. When bound to KRT17, regulates protein synthesis and epithelial cell growth by stimulating Akt/mTOR pathway. May also regulate MDM2 autoubiquitination and degradation and thereby activate p53/TP53.

ab14123 staining mouse embryonic skin tissue sections by IHC-P. Sections were PFA fixed and subjected to heat mediated antigen retrieval in citrate buffer prior to blocking with a commercially available (mouse-on-mouse) blocking reagent for 1 hour at RT. The primary antibody was diluted 1/50 and incubated with the sample for 24 hours at 4°C. A biotinylated mouse anti-mouse antibody was used as the secondary, followed by a Streptavidin-Cy3®.
All lanes : Anti-14-3-3 sigma antibody [1433S01] (ab14123) at 0.5 µg/ml
Lane 1 : U251 (non malignant glioma cells)
Lane 2 : CFBE41o- cells stably transfected with wild-type cystic fibrosis transmembrane conductance regulator (CFTR).
Lane 3 : CFBE41o- cells stably transfected with deltaF508 of cystic fibrosis transmembrane conductance regulator (CFTR).
Lysates/proteins at 10 µg per lane.
All lanes : Goat anti mouse (HRP) at 1/5000 dilution

ab14123 at 1/50 staining human epithelium tissue sections by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). The tissue was paraformaldehyde fixed and a heat mediated antigen retrieval step was performed. The tissue was incubated with the antibody for 30 minutes. An HRP conjugated goat polyclonal antibody was used as the secondary.
ICC/IF image of ab14123 stained SHSY5Y cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab14123, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
