    Abcam’s 17 beta Estradiol in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of 17 beta Estradiol in serum and plasma (citrate).

    A 96-well plate has been precoated with anti-Estradiol IgG. Samples and the Estradiol-HRP conjugate are added to the wells, where Estradiol in the sample competes with the added Estradiol-HRP for antibody binding. After incubation, the wells are washed to remove unbound material and TMB substrate is then added which is catalyzed by HRP to produce blue coloration. The reaction is terminated by addition of Stop Solution which stops the color development and produces a color change from blue to yellow. The intensity of signal is inversely proportional to the amount of Estradiol in the sample and the intensity is measured at 450 nm.

I like these kits from Abcam because they are easy to use and they also have a very good manufacturer's instruction. The kit comes with everything needed (see picture) and the strips wells are very good to maximise the experimental conditions and repeat the assay on a different day. The kit allows to have results in 3 hours, and that is great for teaching in particular. I would definitely recommend this kit.

Human estrogen ELISA to detect mouse estrogen levels.

I attempted this kit twice to determine estrogen levels in mice. The first time many of my samples were out of range so I tried to optimize by leaving my samples overnight and lowering my standard values to get a better estimation of the lower concentrations. However, it appears that the levels of mouse estrogen are too low to be detected by this kit. My highest values were seen in my female mice that underwent an ovariectomy (F ovx samples) 5 weeks prior to testing their plasma - I expected these levels to be nearly zero. While I am sure this kit works well for humans, I do not recommend using it for mice that are not in heat. I have attached my graphed data. Key M: male mice, F: female mice, F (ovx): ovariectomized mice, F sham: female mice that underwent a sham surgery 2 hours means the sample was left for 2 hours, ON means the sample was incubated overnight.

Question

I have serum that gives to high of a signal at full strength. I typically dilute in PBS/1% BSA. Is this dilution buffer compatible with this kit?

Answer

It is recommended to use the kit component 'Estradiol 17-beta Standard 0' to dilute high value samples. The standard is prepared from 17 Beta-Estradiol free serum. Alternatively, serum from males free of Estradiol should be use as the base to dilute high samples. The use of PBS/1% BSA does not mimic the matrix or serum samples and could result in unreliable results.

Question

Customer has kindly contacted us regarding the expected relationship between absorbance and concentration. When running the kit they found this relationship to be inverse (the highest concentration standard had lowest absorbance value). Is this to be expected with this product?
