    Protein tyrosine nitration results in a post-translational modification that is increasingly receiving attention as an important component of nitric oxide signaling. While multiple nonenzymatic mechanisms are known to be capable of producing nitrated tyrosine residues, most tyrosine nitration events involve catalysis by metalloproteins such as myeloperoxidase, eosinophilperoxidase, myoglobin, the cytochrome P-450s, superoxide dismutase and prostacyclin synthase. Various studies have shown that protein tyrosinenitration is limited to specific proteins and that the process is selective. For example, exposure of human surfactant protein A, SP-A, to oxygen-nitrogen intermediates generated by activated alveolar macrophages resulted in specific nitration of SP-A at tyrosines 164 and 166, while addition of 1.2 mMCO 2 resulted in additional nitration at tyrosine 161. The presence of nitrotyrosine-containing proteins has shown high correlation to disease states such as atherosclerosis, Alzheimer’s disease, Parkinson’s disease and amyotrophic lateral sclerosis.

Thank you for contacting us. The note on the nitrotyrosine antibody datasheet stating a predicted molecular weight of 1 kDa is incorrect with regards to what one can expect to see in a western blot. Isolated nitrotyrosine may itself be about 1 kDa but we expect the antibody will detect it as a component of proteins. You will likely see multiple bands of various sizes, depending on how many proteins in your sample contain nitrated tyrosine. For example, the reference at the following link shows, in figure 4, western blot detection of nitrated presenilin after first immunoprecipitating the protein from brain cortex samples.

Thank you for your enquiry. I am sorry to confirm that non of our Oxoguanine 8, Nitrotyrosine, and Malondialdehyde antibodies have been tested with half Karnovsky's fixative. Regrettably, this means we therefore cannot confirm and guarantee how well this fixative would work with these antibodies. I have been provided the following information from our sources: ab64548 Oxoguanine 8 antibody [2Q2311] This antibody has been tested with a)  ice-cold methanol or b)  2% paraformaldehyde followed by ice-cold 70% ethanol.  A brief literature search did not turn up any references that used this clone with half Karnovsky's fixative.   ab78163  Nitrotyrosine antibody [2A8.2] We have not tested this antibody with half Karnovsky's fixative.  Only 10% neutral buffer formalin has been used. ab94883 Nitrotyrosine antibody [HM11] Not tested with Karnovsky's fixative         ab53232 Anti-3-Nitrotyrosine antibody [11C2] Not tested with Karnovsky's fixative    ab6463  Anti-Malondialdehyde antibody For all reference that we have, tissues fixation was done only with paraformaldehyde (4% is a standard concentration). Glutaraldehyde is not necessary. The only antibody I have regrettably not yet been able to obtain more information on is ab61392  Anti-3-Nitrotyrosine antibody [39B6]. I would assume that this has not been tested in Karnovsky's, as this would be quite unusual for IHC testing. However, if I obtain any further details I will send these on to you as soon as possible.
