We purchased two 5-Hydroxymethyl-2’-deoxyuridine antibodies (both ab95259 and ab93155) from Abcam but none of them works on dot blot. We tried multiple conditions and there was simply no signal, contrary to what's posted on line (ab93155 is said to work on WB). We are very disappointed about these products from Abcam. We notice that there is a third one (https://www.abcam.com/5-Hydroxymethyluridine-antibody-ab19735.html) that is said to work on ELISA. We are planning to order this Ab to give a final shot. But I can not persuade our supervisor to purchase the ab19735 Ab since he is very upset about the product from Abcam. I'm wondering if you could send me a small aliquot of the ab19735 Ab to test. If it works well, I'm sure he will consider purchasing it immediately.

While Dot Blot does indeed have a lot of similarity to a non-reduced/denatured western blot or with an indirect ELISA, we do consider it to be a separate application from either of these. We do however recognize that many products have not been tested for use in this application and as such using a product which is guaranteed for use in one of these is probably the best bet when searching for antibodies.
If you could provide me with the details of your protocol and with any troubleshooting steps and images which you may have, I would be very happy to review these and see if I might be able to provide suggestions to improve the performance of these products.While these are not guaranteed for use in Dot Blot, should we not be able to provide help with these I am sure we might be able to find a way to come to a result which would be satisfactory to both Abcam and you.
