Hi John   (Anti 5-mC query) The antigen retrieval that was performed was 15 mins in the pressure cooker in citrate buffer pH 6 then on the bench for 20 minutes. I have advised her to leave for longer in the pressure cooker. If there’s anything else you or your colleagues can suggest I would be grateful.   She won’t have time to repeat the staining for another 2 weeks, if it still doesn’t work after that we could try another batch of antibody.  


Thanks you for sending the information on you IHC protocol. I am still trying to see if I can find any IHC protocols that are better than others for detected DNA modifications. If I find any I will send them on as soon as I find them. In the meantime, I have attached our general IHC guide, which contains information on several antigen retrieval methods.
