Asynchronous cells were paraformaldehyde-fixed, permeablized, immunofluorescently labeled and counterstained with DAPI. This antibody produces a weak staining pattern that is only slightly enriched within nucle - it appears to be predominantly non-specific staining. MeOH-fixed samples were also evaluated and failed to produce the expected staining pattern.
This antibody has been successfully used for ICC/IF under different experimental conditions. The fact that you have not been able to get a good result with this antibody might imply that not all protocols will be equally suitable and that the results obtained are highly dependent on the method used.
