    The native context of double-stranded DNA may obstruct antibody binding to 5-methylcytosine. For successful detection of 5-methylcytosine, we recommend that the DNA is denatured to make the nucleotides accessible for the antibody. Denaturing methods vary depending on each application.

Preparation
Cell harvesting/tissue preparation method: For the extraction, mice are intraperitoneally injected with around 3mL of sterile Hank's medium at 37¹C and gently massaging the abdomen we recover the volume that exits from the aperture, this procedure allows the mice to live after extraction. Then, to remove adherent macrophages and B lymphocytes, that volume is placed on a MIF plate (Kartell Labware) for 30min and the supernatant enriched in T lymphocytes is recovered.
