Target

  • Function

    This is one of the several different receptors for 5-hydroxytryptamine (serotonin), a biogenic hormone that functions as a neurotransmitter, a hormone, and a mitogen. This receptor is a ligand-gated ion channel, which when activated causes fast, depolarizing responses in neurons. It is a cation-specific, but otherwise relatively nonselective, ion channel.
  • Tissue specificity

    Expressed in cerebral cortex, amygdala, hippocampus, and testis. Detected in monocytes of the spleen and tonsil, in small and large intestine, uterus, prostate, ovary and placenta.
  • Sequence similarities

    Belongs to the ligand-gated ion channel (TC 1.A.9) family. 5-hydroxytryptamine receptor (TC 1.A.9.2) subfamily. HTR3A sub-subfamily.

    IHC image of ab13897 staining in human normal cerebral cortex formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab13897, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

The amino acids correspond to the protein sequence with the signal peptide (SwissProt: P35563). We normally look for at least 85% degree of homology before we would recommend using an antibody in an untested species. I cannot find the Xenopus sequence, but if you would like to send it to me, i will be happy to do the alignment and see if there is a good chance that the antibody would work.

Thank you for contacting us. The detection kit we used for the IHC image is the Leica Bond refine polymer detection kit (for the automated Leica Bond IHC system). The system uses small linkers to enhance and refine the detection of the target protein, linked through a compact polymer backbone. The kit contains a peroxide block, a post primary, polymer, DAB chromogen and a haematoxylin counterstain. I hope this information is helpful to you.
